This study describes for the first time an abortion outbreak caused by Neospora caninum in farmed red deer. During a 5-year period, farmed hinds, naturally mated, were regularly ultrasound monitored to detect reproductive losses over their gestation. During the 4 years previous to the outbreak, abortion rates ranged from 4.7 to 8.6% (average 6.5%), and serology for indirect diagnosis of neosporosis and toxoplasmosis was performed. At the fifth year, the abortion rate increased to 25.3%. During this outbreak, three aborted foetuses and their placentas were recovered and submitted to laboratory for etiological diagnosis. Blood samples were collected from the 81 hinds at the end of the gestational period and the seropositivity rate for N. caninum, Toxoplasma gondii, Brucella abortus, bovine viral diarrhoea virus and bovine alphaherpesvirus type 1 was 66.7%, 67.9%, 0.0%, 8.6% and 0.0%, respectively. Neospora caninum-seropositive hinds (OR = 5.7, P = 0.0271) and hinds with high antibody titres to N. caninum (OR = 7.4, P = 0.0130) were more likely to abort than seronegative hinds. In addition, N. caninum seropositivity rate in the aborted hinds was higher (OR = 5.4, P = 0.033) than the non-aborted hinds. No association was found between T. gondii nor BVDV-seropositivity and abortions. Typical protozoal histopathologic findings (necrotizing non suppurative encephalitis, meningitis, myocarditis, hepatitis, among others) were observed in all foetuses. Neospora caninum was immunolabelled by immunohistochemistry in several tissues from two foetuses, and infection was also confirmed in the three foetuses by serology and/or DNA detection. No other abortifacient agent was detected in the foetuses. Their dams showed high N. caninum antibody titres (≥ 6400). Serologic evidence and epidemiological data recorded suggested a point-source of N. caninum infection before the occurrence of the outbreak, probably related with contaminated feedstuff with oocysts. Moreover, the intensive production system with a high stocking rate could be also considered a factor which might have increased the risk of horizontal N. caninum infection in this herd.

Peste des Petits Ruminants Virus (PPRV) is an extremely infective morbillivirus that primarily affects goats and sheep. In underdeveloped countries where livestock are the main economical resource, PPRV causes considerable economic losses. Protective live attenuated vaccines are currently available but they induce antibody responses similar to those produced in PPRV naturally infected animals. Effective vaccines able to distinguish between vaccinated and naturally infected animals are required to PPRV control and eradication programs. Hemagglutinin (H) is a highly immunogenic PPRV envelope glycoprotein displaying both hemagglutinin and neuraminidase activities, playing a crucial role in virus attachment and penetration. In this study, a recombinant Bovine Herpesvirus-4 (BoHV-4)-based vector delivering an optimized PPRV-Hemagglutinin expression cassette, BoHV-4-A-PPRV-H-ΔTK, was assessed in immunocompetent C57BL/6 mice. BoHV-4-A-PPRV-H-ΔTK-immunization elicited both cellular and humoral immune responses with specific T cell, cytotoxic T lymphocyte, and sero-neutralizing antibody against PPRV. These data suggest recombinant BoHV-4-A-PPRV-H-ΔTK as an effective vaccine candidate to protect against PPRV herd infection and potentially applicable for eradication programs.

Bovine herpesvirus 4 (BoHV-4) is a promising vector for the delivery and intracellular expression of recombinant antigens and can thus be considered as a new prototype vaccine formulation system. An interesting, and actively pursued, antigen in the context of human immunodeficiency virus (HIV) infection prophylaxis (and therapy) is the C-C chemokine receptor type 5 (CCR5) co-receptor, whose blockage by specific antibodies has been shown to inhibit both viral entry and cell-to-cell transmission of the virus. Building on our previous work on the BoHV-4 vector system, we have engineered and tested a replication-competent derivative of BoHV-4 (BoHV-4-CMV-hCCR5ΔTK) bearing a human CCR5 (hCCR5) expression cassette. We show here that CCR5 is indeed expressed at high levels in multiple types of BoHV-4-CMV-hCCR5ΔTK-infected cells. More importantly, two intravenous inoculations of CCR5-expressing BoHV-4 virions into rabbits led to the production of anti-CCR5 antibodies capable of reacting with the CCR5 receptor exposed on the surface of HEK293T cells through specific recognition of the amino-terminal region (aa 14-34) of the protein. Given the growing interest for anti-CCR5 immunization as an HIV control strategy and the many advantages of virus-based immunogen formulations (especially for poorly immunogenic or self-antigens), the results reported in this study provide preliminary validation of BoHV-4 as a safe viral vector suitable for CCR5 vaccination.

Idiopathic pulmonary fibrosis is a very common interstitial lung disease derived from chronic inflammatory insults, characterized by massive scar tissue deposition that causes the progressive loss of lung function and subsequent death for respiratory failure. Bleomycin is used as the standard agent to induce experimental pulmonary fibrosis in animal models for the study of its pathogenesis. However, to visualize the establishment of lung fibrosis after treatment, the animal sacrifice is necessary. Thus, the aim of this study was to avoid this limitation by using an innovative approach based on a double bleomycin treatment protocol, along with the in vivo images analysis of bleomycin-treated mice. A reporter gene construct, containing the luciferase open reading frame under the matrix metalloproteinase-1 promoter control region, was tested on double bleomycin-treated mice to investigate, in real time, the correlation between bleomycin treatment, inflammation, tissue remodeling and fibrosis. Bioluminescence emitted by the lungs of bleomycin-treated mice, corroborated by fluorescent molecular tomography, successfully allowed real time monitoring of fibrosis establishment. The reporter gene technology experienced in this work could represent an advanced functional approach for real time non-invasive assessment of disease evolution during therapy, in a reliable and translational living animal model.

The pathogenic role of BoHV-4 remains unclear and it is unknown whether the virus acts as a primary pathogen or whether it facilitates secondary infections After natural or experimental infections, BoHV-4 can establish latency, mainly in cells of the monocyte/macrophage linage. Latent virus can be reactivated after glucocorticoid treatment or by stress factors. In 2007, BoHV-4 was isolated for the first time in Argentina, from samples of bovine abortions. In the present study, we used viral isolation, nested PCR and restriction endonuclease analysis (REA) to investigate the presence of BoHV-4 in bovine leukocytes from a single herd of dairy cattle with reproductive problems. In this work, we demonstrated that BoHV-4 genome is present in the leukocytes of a high proportion (63.4%) of animals, probably in a latent or persistent state. BoHV-4 was isolated from one out of eleven peripheral blood leukocyte (PBL) samples. By REA we demonstrated the existence of genomic variation among the strains circulating in this particular herd. Furthermore, all PBL samples evaluated in this study differed from the American prototype strain, DN 599. Overall, this work demonstrated that BoHV-4 is present in the leukocyte fraction of dairy cattle and that viral strains present in this herd are genetically divergent. Although BoHV-4 was detected in a herd with a background of reproductive disorders, it is not possible to conclude that the virus is the primary responsible for these conditions.


O herpesvírus bovino tipo 4 (BoHV-4) é um gama-herpesvírus que foi isolado de animais aparentemente saudáveis e de gado com uma variedade de sinais clínicos. O papel patogênico do BoHV-4 ainda não está claro e não se sabe se o vírus age como um patógeno primário ou se facilita infecções secundárias. Depois de infecções naturais ou experimentais, BoHV-4 pode estabelecer latência, principalmente nas células dos linhagens de monócitos/macrófagos. O vírus latente pode ser reativado após o uso de glicocorticóides ou por fatores de estresse. Em 2007, o BoHV-4 foi isolado pela primeira vez na Argentina, a partir de amostras de abortos bovinos. No presente estudo, utilizou-se o isolamento viral, nested PCR e análise com endonucleases de restrição (REA) para investigar a presença de BoHV4 em leucócitos de bovinos provenientes de um único rebanho de gado leiteiro com problemas reprodutivos. Neste trabalho, demonstramos que o genoma do BoHV-4 está presente nos leucócitos em uma elevada proporção (63,4%) dos animais, provavelmente em um estado latente ou persistente. BoHV-4 foi isolado de uma de cada onze amostras de leucócitos no sangue periférico (PBL). Por REA nós demonstramos a existência de variações genômicas entre as estirpes circulantes deste rebanho particular. Além disso, todas as amostras de PBL avaliados neste estudo diferiram da estirpe protótipo Americano, DN 599. Em geral, este estudo demonstrou que o BoHV-4 está presente na fração leucocitária do gado leiteiro e que as estirpes virais presentes neste rebanho são geneticamente divergentes. Embora que BoHV-4 foi detectado em um rebanho com história de distúrbios reprodutivos, não é possível concluir que o vírus é o principal responsável por estas condições.
